TNFR2 expression is a hallmark of human memory B cells with suppressive function

چکیده

Tumor Necrosis Factor Receptor 2 (TNFR2) expression is increasingly being linked to tolerogenic immune reactions and cells with suppressor function including a subset of T-regulatory cells. B-regulatory play an important role in control T-cell responses inflammation. Recently, we described TNFR2 as marker for IL-10-producing B cells, hallmark this cell subset. Here, demonstrate that proliferation T reduced the presence positive human memory generated TLR9 ligand, while TNFR2- TNFR2+CD27- display costimulatory activity. Our data further reveal IL-10 secretion characteristic IgM+ naïve but suppressive activity not restricted IL-10: (i) inhibitory effect TNFR2+ switched was comparable exerted by although levels cocultures were lower; (ii) supernatants from failed suppress proliferation. Based on our findings, propose formation Breg specific undergoing terminal differentiation. corroborate represents viable identification regulatory function.

Myeloid suppressor cells require membrane TNFR2 expression for suppressive activity

TNF and TNF receptor type 2 (TNFR2) have been shown to be important for generation of myeloid-derived suppressor cells (MDSC). In order to analyze whether and how TNFR2 passes the effect of TNF on, myeloid cells from TNFR2-deficient mice were compared to respective cells from wild-type mice. Primary TNFR2-deficient myeloid cells showed reduced production of NO and IL-6 which was attributable to...

CD127 expression inversely correlates with FoxP3 and suppressive function of human CD4+ T reg cells

Regulatory T (T reg) cells are critical regulators of immune tolerance. Most T reg cells are defined based on expression of CD4, CD25, and the transcription factor, FoxP3. However, these markers have proven problematic for uniquely defining this specialized T cell subset in humans. We found that the IL-7 receptor (CD127) is down-regulated on a subset of CD4(+) T cells in peripheral blood. We de...

Human IgM+CD27+ B cells: memory B cells or "memory" B cells?

Memory B cells are generated in germinal centers (GC) and contribute to serological immunity by rapidly differentiating into plasma cells. Human memory B cells can be identified by the expression of CD27. These cells exhibit more rapid responses than naive (CD27-) B cells following stimulation in vitro, consistent with the heightened kinetics of secondary responses in vivo. CD27+ B cells expres...
